I suggest:

1) New toolbox etc gui
2) Colorspace independence including CMYK and at least one polar coordinate 
colorspace (lab or luv comes to mind, though hsv is more commonly known)

With that done then playing with those funny and interesting 
colorspaces/paintmodels can be done again, though they'll probably not be 
ready for the next version

Also a lot of smaller improvements here and there.

Just my thoughts though
