Hi,

unless there's a direct fly to Brno, the best option is to fly to Vienna or 
Prague and take a coach from there.

The coach from Vienna departs directly from the terminal at Vienna Airport. It 
goes every 2 hours, takes approximately 2 and half hour and it's about €13. 
This is the best option, since you can't possibly get lost :)

The coach from Prague to Brno takes the same time and costs €9. It goes every 
30 minutes. You need to get from the airport to the bus station though, which 
can take up to one hour.

It's also possible to fly to Bratislava and take a coach from there, but I 
have no experience with it.

All the bus lines are run by Student Agency (http://www.studentagency.eu/, not  
just for students ;-) ) with very modern and comfortable buses where you get a 
free hot beverages, newspaper and they have free wifi.
