When you insert such a device, the encrypted parition appears along side any 
unencrypted on in the device notifier. Clicking the padlock icon on the right 
hand side shows a password dialog and then unlocks the device (provided the 
input is correct of course).

Locking again is just a matter of another click on the padlock.
